BRAF gene mutations represent a key oncogenic alteration across a range of solid tumors and, at the same time, a significant therapeutic target in the era of precision oncology. This article aims to provide an overview of current options for targeted therapy of BRAF-mutated tumors and key clinical data. Based on the results of melanoma studies demonstrating the high efficacy of combined BRAF and MEK inhibition, and subsequent basket studies confirming activity across various diagnoses, BRAF has established itself as a model example of a tumor-agnostic therapeutic approach. At the same time, differences in treatment efficacy depending on the biological context of individual tumors are discussed, particularly in colorectal cancer, where a combination strategy beyond BRAF inhibition is often necessary. A review of available data shows that targeted BRAF inhibition is associated with significant antitumor activity and represents an important component of modern systemic therapy across tumor indications.

Keywords: BRAF oncogene, BRAF V600E mutation, tumor-agnostic treatment.
